Thymosin alpha-1 is a synthetic 28-amino-acid peptide whose sequence was first identified in extracts of bovine thymus tissue during the 1970s. The chain carries an acetyl group on its N-terminal serine. Its acidic residue content is high, which produces strong water solubility and an isoelectric point well below neutrality. Material supplied for laboratory and clinical use is manufactured by solid-phase peptide synthesis rather than purified from animal tissue. Different salt forms, such as the acetate, alter the counter-ion content without changing the peptide backbone.
Whether the free 28-residue peptide circulates in human tissue remains debated. The best-documented human source is prothymosin alpha, a larger acidic protein that carries the sequence at its N-terminus. Reports of measurable peptide levels in serum and lymphoid tissue exist, yet some of that signal may come from cross-reacting fragments or from the parent protein. Most reviews therefore treat prothymosin alpha as the established human molecule and describe independent circulation of the small peptide as an unresolved question.
Long-term storage is generally at minus twenty degrees Celsius or colder, preferably desiccated and protected from light. Lyophilized material is more stable than reconstituted solution, which degrades faster at room temperature. Stability depends on pH, ionic strength, and the presence of oxidising agents. Published stability data for the peptide are limited, so storage claims in catalogues should be treated as general guidance rather than measured guarantees. Freeze-thaw cycles are kept to a minimum.
Identity and purity are normally confirmed by reversed-phase high-performance liquid chromatography and mass spectrometry. The expected mass for the acetylated 28-residue peptide is close to 3108 daltons, and a mass shift indicates a modification or truncation. Peptide mapping after enzymatic digestion can resolve sequence-level questions. Counter-ion content, water content, and residual solvents are separate quality attributes that a certificate of analysis may or may not report. Aggregation is monitored by size-exclusion chromatography when relevant.
| Property | Value | Notes |
|---|---|---|
| Amino acid length | 28 residues | Single chain with an acetylated N-terminus |
| Molecular mass | Approximately 3,108 Da | Small shifts occur with counter-ion and water content |
| Isoelectric point | Around 3.5 | Low value follows from the many acidic residues |
| Parent molecule | N-terminal region of prothymosin alpha | Free circulating form in humans is not firmly established |
| Common synonyms | Thymalfasin; T alpha 1 | Older literature also uses the full spelled-out form |
Identity and purity are usually assessed by reversed-phase high-performance liquid chromatography, which separates the target peptide from truncated or chemically modified byproducts. Mass spectrometry confirms the expected molecular mass and can indicate acetylation state or sequence errors. Amino acid analysis and peptide mapping supply complementary sequence-level information, while endotoxin testing is relevant for preparations intended for cell or animal work. Purity figures reported by suppliers refer to the method used and are not directly comparable across laboratories unless conditions are stated.
Lyophilized material is typically treated as a hygroscopic solid that should be brought to room temperature before the container is opened, which limits condensation on the powder. Reconstitution is commonly done with sterile water or a buffered diluent, and gentle mixing is preferred over vigorous agitation to reduce foaming and surface adsorption. Because peptides can bind to plastic and glass, diluents containing a small amount of carrier protein are sometimes used in laboratory work. Working solutions are generally aliquoted and prepared fresh rather than subjected to repeated freezing and thawing.
Stability depends on temperature, pH, and the number of freeze-thaw events the sample has experienced. Freeze-dried material is commonly held at -20 °C or colder, while reconstituted liquid is kept cold and used within a short window. Extreme pH and prolonged light exposure can promote deamidation, oxidation, or aggregation, particularly at asparagine and methionine positions. Adsorption to container walls can lower the measured concentration of a dilute solution even when the peptide molecules themselves remain intact.
Recommended storage for the dry powder is a freezer near minus twenty degrees Celsius, kept desiccated and away from light. Once dissolved, the peptide is less stable and is usually held at two to eight degrees Celsius for short intervals or frozen for longer storage. Stability studies focus on the acetylated terminus and the disulfide linkage because those features define the intact molecule. Common degradation routes include cysteine oxidation, deamidation of asparagine or glutamine side chains, and slow formation of higher-molecular-weight species.
Identity and purity are usually checked by reverse-phase high-performance liquid chromatography, which separates the intact chain from truncated products, together with mass spectrometry for confirmation of the expected mass. Peptide mapping after enzymatic digestion and amino acid analysis add sequence-level evidence. Release testing also covers water content, residual solvents, and counter-ions, all of which influence measured mass and stability. Related-peptide limits are commonly expressed as a percentage of total peak area, with individual unspecified impurities held below a lower threshold.

== Tissue and subcellular distribution == Galectin-7 is found both intracellularly and extracellularly. It is secreted via a non-classical, endoplasmic reticulum–Golgi–independent pathway and can form galectin–glycan lattices by clustering on cell-surface receptors. It is also present in primary cilia of epithelial cell types such as those of the airway and kidney.
=== First confirmed cases: May–June 2022 === On 12 May 2022, the North Korean government declared a "severe national emergency", after samples from an unspecified number of people tested positive for COVID-19. This marked the first time that North Korea had publicly acknowledged the existence of COVID-19 cases in the country. The Korean Central News Agency stated that Supreme Leader Kim Jong Un had called an emergency meeting of the Politburo of the Workers' Party of Korea after learning of the samples, which were sourced from residents of Pyongyang and had symptoms "consistent with" the Omicron variant. The Politburo recommended the implementation of a "maximum" emergency quarantine, to include nationwide lockdowns, border restrictions, and restrictions on group sizes in workplaces. During the politburo meeting, the previous anti-pandemic strategy was criticised. NK News reported that the entire country had been placed under a lockdown two days prior, though farmers in border regions close to South Korea were seen still tending the fields. It was later reported by state media that at least one North Korean died after testing positive, and that 187,800 people are now under quarantine due to "fever". On 13 May, Kim Jong Un held a meeting at the State Emergency Epidemic Prevention Headquarters, where he called for further anti-pandemic work, lockdowns, and isolation of suspected cases. The effort to stop the spread was declared to be a supreme task of the party.
Unpressed bittern-gelled soft tofu is called sun-dubu (순두부; "mild tofu") in Korean. Soy milk is mixed with seawater, or saline water made with sea salt, so that it curdles. The curds remain loose and soft. Freshly made sun-dubu is eaten boiled with little or no seasoning. Manufactured sundubu is usually sold in tubes. It is also the main ingredient in sundubu-jjigae (순두부찌개; "soft tofu stew"). Although the word sun in sun-dubu does not have a Sino-Korean origin, sun-dubu is often translated into Chinese and Japanese using the Chinese character 純, whose Korean pronunciation is sun and the meaning is "pure". Thus in China, sun-dubu is called chún dòufu (純豆腐; "pure tofu"), and in Japan, it is called jun-tōfu (純豆腐) or sundubu (スンドゥブ).

=== Growth hormone receptor mutations === Molecular genetic investigations have shown that LS is mainly associated with autosomal recessive mutations in the gene for the growth hormone receptor (GHR). These can result in defective hormone binding to the ectodomain or reduced efficiency of dimerization of the receptor after hormone occupancy. LS is generally classified as "primary" GH insensitivity, which is distinguished from "secondary" GH insensitivity. Primary (congenital/hereditary) GH insensitivity may result from growth hormone receptor defects, as in the case of Laron syndrome, but can also be caused by defective post-receptor signal transduction (STAT5B), abnormalities of the IGF-1 gene or IGF-1 receptor. Secondary (acquired) GH insensitivity results from antibodies to growth hormone or the growth hormone receptor, as well as poor nutritional status, liver disease or diabetes mellitus. A GHR mutation that results in only partial insensitivity to GH can manifest as a form of idiopathic short stature.
To what extent suicidal thoughts are influenced by behavioral symptoms is unclear, as they signify a desire to avoid the later stages of the disease. Suicide is the greatest risk of this disease before the diagnosis is made and in the middle stages of development throughout the disease. Other associated risks include choking due to the inability to swallow, physical injury from falls, and malnutrition.
== Terminology == Desmoplasia originates from the Ancient Greek δεσμός desmos, 'knot, bond' and πλάσις plasis, 'formation'. It is usually used in the description of desmoplastic small round cell tumors. Neoplasia is the medical term used for both benign and malignant tumors, or any abnormal, excessive, uncoordinated, and autonomous cellular or tissue growth.
== Treatment == The main focus of treatment for AGAT deficiency is supplementation of creatine, with the goal of replenishing cerebral creatine to normal levels. This is done with oral creatine supplementation. Treatment is most effective if it is started early in life, before symptoms are apparent. Treatment in affected individuals does not reverse intellectual disability or improve cognitive function. For treatment at any age, even if intellectual disability was present, all individuals showed improvement in muscle weakness. In an asymptomatic sibling, who was started on treatment due to the earlier diagnosis of an affected sibling, early intervention with creatine supplementation resulted in improved outcomes when compared to their untreated siblings at the same age. In addition to clinical findings, the effectiveness of treatment can be monitored by following creatine levels in blood and urine as well as the creatine signal by MRS. As creatine is rapidly converted and excreted as creatinine, treatment must be life long to continue to benefit the patient. Treatment during the early years of brain development is most important for preserving brain function.

== Gene expression == This gene encodes a member of the peroxisome proliferator-activated receptor (PPAR) subfamily of nuclear receptors. PPARs form heterodimers with retinoid X receptors (RXRs) and these heterodimers regulate transcription of various genes. Three subtypes of PPARs are known: PPAR-alpha, PPAR-delta, and PPAR-gamma. The protein encoded by this gene is PPAR-gamma and is a regulator of adipocyte differentiation. Alternatively spliced transcript variants that encode different isoforms have been described. The activity of PPARG can be regulated via phosphorylation through the MEK/ERK pathway. This modification decreases transcriptional activity of PPARG and leads to diabetic gene modifications, and results in insulin insensitivity. For example, the phosphorylation of serine 112 will inhibit PPARG function, and enhance adipogenic potential of fibroblasts.

While the amount of radiation from the plugs was minuscule and not a threat to the consumer, the benefits of such plugs quickly diminished after approximately a month because of polonium's short half-life and because buildup on the conductors would block the radiation that improved engine performance. (The premise behind the polonium spark plug, as well as Alfred Matthew Hubbard's prototype radium plug that preceded it, was that the radiation would improve ionization of the fuel in the cylinder and thus allow the motor to fire more quickly and efficiently.)

It is usually described as an immunomodulatory peptide rather than a classic circulating hormone. No endocrine gland is known to release it as a primary secretory product, and its measured presence in blood is not firmly established.
Prothymosin alpha is a much larger acidic protein, roughly 111 to 113 residues long, and the thymosin alpha-1 sequence matches its N-terminal region. The small peptide is therefore best understood as a fragment of that parent protein rather than a separate gene product.
Trials and clinical reports have examined chronic hepatitis B and C, use as a vaccine adjuvant, and supportive treatment in some immunodeficiency and oncology settings. Results vary by indication, and regulatory approval differs between countries.
Lyophilized powder is normally kept at minus twenty degrees Celsius or below, in a sealed container, desiccated and away from light. Reconstituted solutions are less stable and are usually refrigerated and used quickly. Repeated temperature cycling should be avoided.
